Avaliação da DL50 e edema pulmonar induzido pelo veneno de Tityus serrulatus (Scorpiones; Buthidae) procedente da Bahia, Brasil Biota Neotropica

O escorpião Tityus serrulatus é conhecido como a espécie mais importante, do ponto de vista médico, pois é o que causa os acidentes mais graves registrados para o território brasileiro. Este trabalho trata da caracterização do veneno do escorpião T. serrulatus, através da obtenção da DL50 e determinação da capacidade em induzir edema pulmonar, em ratos. O veneno foi obtido através de estímulo elétrico. A toxicidade foi determinada através da avaliação da DL50, pelo método de Finney (1971). Os valores demonstraram a baixa toxicidade do veneno (96,16 mg/camundongo), que corresponde a 3 - 7 vezes menos tóxico que venenos de espécimes de outras regiões do Brasil. Accidental envenomation caused by Tityus serrulatus scorpions is very common in Brazil and may result in serious cardiorespiratory alterations that are frequently fatal to children. In the present study, the effects of T. serrulatus venom on the cardiorespiratory system of recently weaned male Wistar rats were evaluated. Fifteen animals were distributed into three groups (n = 5). Scorpionism is a common problem that occurs in tropical and subtropical countries and assumes great medical-sanitary importance due to its fatal effect on sensitive individuals, being able to lead children and aged people to death. Convulsive effects of some isolated venom fractions of the Tityus serrulatus scorpion: behavioral, electroencephalographic, and neuropathological aspects J. Venom. Anim. Toxins

It has been previously shown that the crude venom of Tityus serrulatus can cause convulsions. This study was designed to investigate the neurotoxic effects of B, C, G, and K fractions isolated from this venom. Intravenous injection of these fractions in mice (0.6 - 6.0 mg/kg body weight) showed that the C fraction is a potent convulsant and G fraction decreased the threshold for tonic hand limb extension elicited by transauricular electroshock. Scorpion venom neurotoxins are responsible for toxicity and pharmacological effects. They are active in sodium and potassium channels leading to an increase in the release of neurotransmitters, such as glutamate. Glutamate is found in large quantities in the hippocampus (HPC) and is involved in the long-term potentiation (LTP) induction. The HPC is known to be related to certain kinds of memory. Tityus serrulatus is the most venomous scorpion in Brazil; however, it is not known whether its venom causes any harm to the offspring whose mothers have received it. This study investigates whether the venom of T. serrulatus may lead to deleterious effects in the offspring, when once administered to pregnant rats at a dose that causes moderate envenomation (3mg/kg). The venom effects were studied on the 5th and on the 10th gestation day (GD5 and GD10). The maternal reproductive parameters of the group that received the venom on GD5 showed no alteration. The hematological and biochemical profiles of newly weaned rats submitted to experimental poisoning with T. serrulatus venom were evaluated. Fifteen recently weaned male Wistar rats (mean weight 130g) were distributed into three equal groups (n = 5). Avaliou-se o hemograma de 12 cães adultos, saudáveis (14,2±5,4kg) após a inoculação de veneno do escorpião amarelo (Tityus serrulatus). Os animais foram distribuídos em dois grupos (G), com seis em cada: os do GI foram usados como controle e receberam 0,5mL de salina tamponada com fosfato (PBS) por via subcutânea (SC) na face medial da coxa esquerda (FMCE), e os do GII receberam veneno liofilizado do T. serrulatus (250µg/kg) diluído em PBS por via SC na FMCE. Induction of neutralizing antibodies in mice immunized with scorpion toxins detoxified by liposomal entrapment BJMBR
The possibility of producing neutralizing antibodies against the lethal effects of scorpion toxins was evaluated in the mouse model by immunization with an immunogen devoid of toxicity. A toxic fraction (5 mg) from the venom of the scorpion Tityus serrulatus was entrapped in sphingomyelin-cholesterol liposomes. The liposomes were treated for 1 h at 37oC with a 1% (w/w) trypsin solution in 0.2 M sodium carbonate buffer, pH 8.3. This treatment led to a strong reduction in venom toxicity. O escorpião amarelo Tityus serrulatus é registrado pela primeira vez para oeste do Rio Grande do Sul. Foram coletados dois espécimes no município de Uruguaiana (29° 45' 23'' S e 57° 05' 37'' W) em junho de 2006. A ocorrência dos espécimes de T. serrulatus no Rio Grande Sul representa um aumento de 2.127 km em sua área de distribuição conhecida.

It has been proposed that the subgenus Archaeotityus comprises the most ancient species group within the medically important scorpion genus Tityus. cDNA encoding sodium-channel active toxins from the type species of this subgenus, Tityus clathratus (central Venezuela), have been isolated and sequenced. Two cDNAs were retrieved that encoded 61 amino acid-long putative neurotoxins named Tcl1 and Tcl2. Selection of environmental temperature by the yellow scorpion Tityus serrulatus Lutz & Mello, 1922 (Scorpiones, Buthidae) J. Venom. Anim. Toxins incl. Trop. Dis.

The preferred temperature of the yellow scorpion Tityus serrulatus was investigated since its dispersion is a matter of concern. Adult T. serrulatus, weighing 1.24 + 0.20 g (mean + sd) and with a standard length of 59.3 + 2.5 mm, were used. A metallic corridor (120 cm long, 5 cm large and 10 cm high) with thermal gradient ranging from 0°C to 40°C was used. Tityus serrulatus chose and stayed in temperatures ranging from 14°C to 38°C when safe conditions were offered (dark and thigmotactic stimuli). The number of animals that remained in the 11°C-20°C, 21°C-30°C, and 31°C-40°C temperature zones were 8, 8, and 9, respectively. In this paper further information concerning the ecological and biogeographical aspects of scorpionism is presented. Some of the information already outlined in three previous publications(10,14,16) is assessed, and new data given concerning the pattern of distribution presented by Tityus serrulatus in Brazil. Some new ideas are also proposed regarding the possible evolutionary advantages and disadvantages of parthenogenesis versus sexuality in both the short-term and the long-term.

Abstract Background In Brazil, accidents with venomous animals are considered a public health problem. Tityus serrulatus (Ts), popularly known as the yellow scorpion, is most frequently responsible for the severe accidents in the country. Ts envenoming can cause several signs and symptoms classified according to their clinical manifestations as mild, moderate or severe. Furthermore, the victims usually present biochemical alterations, including hyperglycemia. Nevertheless, Ts envenoming and its induced hyperglycemia were never studied or documented in a patient with diabetes mellitus (DM).
